    1. Vigene生物购买的人类ORF开放阅读框克隆的序列与参考序列完全一致吗?

    克隆序列并不总是与RefSeq的公布的序列完全一致。Vigene生物的大部分人类ORF克隆的序列与NCBI公布的参考序列完全匹配,但部分克隆的序列含有SNP单核苷酸多态性,存在短序列的插入或缺失。Vigene生物人类ORF的亚克隆模板来自哺乳动物MGC库或cDNA文库,序列的差别反映了来自不同组织和不同个体的差异。

     

    2. Vigene生物所有的人类ORF克隆都已全长测序验证了吗?

    是的。Vigene生物科技有限公司所有的人类ORF克隆都已通过全长序列测序。每个克隆首先用Sanger法对两端序列进行测序验证,然后通过NextGene二代测序方法对全长序列测序。每个克隆的测序结果已在我们的官方网站www.vigenebio.cn上公布。Vigene克隆的序列与参考序列之间的差异也可在网站上查阅。提交您的订单前,我们强烈建议您下载我们的序列并比对差异。

    3. 在什么情况下可以选用腺病毒载体作为将外源基因导入细胞的工具?

    腺病毒载体能感染分裂和非分裂细胞。它适用于体内和体外基因传递,转染效率高,对外源基因瞬时表达量高。但相对于其他病毒载体系统,腺病毒载体在靶细胞中具有相对较高的免疫原性。

     

    您应考虑以下几点: 

    1. 外源基因需要瞬时表达还是稳定的基因表达?

    2. 需转染分裂细胞还是非分裂的细胞?

    3.对于靶细胞的潜在免疫原性是否有充分的了解?
